Venkatraman Ramakrishnan wins Nobel Prize chemistry 2009

Venkatraman Ramakrishnan Stockholm, Oct 7 (THAINDIAN NEWS) Venkatraman Ramakrishnan, Thomas A Steitz and Ada E Yonath have won this year’s Nobel Prize in Chemistry, it was announced today.

Venkatraman Ramakrishnan is an Indian-American. And he shared the award with two other scientists for their path-breaking work on ribosomes, which may help in the development of new antibiotics.

The 57-year-old Ramakrishnan, who belongs to Chidambaram in Tamil Nadu, will share USD 1.4 million prize money with Thomas Steitz of the US and Ada Yonath of Israel.

The Royal Swedish Academy of Sciences said three-dimensional models developed by the three scientists showed how different antibiotics bind to ribosomes. 

”These models are now used by scientists in order to develop new antibiotics, directly assisting the saving of lives and decreasing humanity’s suffering,” the academy said in its citation.

Venkatraman Ramakrishnan is the 7th Indian to win the Nobel Prize, after Rabindranath Tagore, Chandrasekhara Venkata Raman, Hargobind Khorana, Mother Teresa, Subramanian Chandrasekhar and Amartya Sen.

Venkatraman is a senior scientist at the MRC Laboratory of Molecular Biology at Cambridge. All the three Nobel laureates for chemistry have used a method called X-ray crystallography to map the position for each and every one of the hundreds of thousands of atoms that make up the ribosome, the citation said.

    A new multisensor image fusion algorithm based on fuzzy logic is proposed. The membership function and fuzzy rules of the new algorithm is defined using the Fuzzy Inference System (FIS) editor of fuzzy logic toolbox in Matlab 6.1. The new algorithm is applied to fuse Charge-Coupled Device (CCD) and Synthetic Aperture Rader (SAR) images. The fusion result is compared with some other fusion algorithms through some performance evaluation measures for the fusion effect, and the comparison results show that the new algorithm is effective.
    An image fusion algorithm, based upon spectral mixture analysis, is presented. The algorithm combines low spatial resolution multi/hyperspectral data with high spatial resolution sharpening image(s) to create high resolution material maps. Spectral (un)mixing estimates the percentage of each material (called endmembers) within each low resolution pixel. The outputs of unmixing are endmember fraction images (material maps) at the spatial resolution of the multispectral system. This research includes developing an improved unmixing algorithm based upon stepwise regression. In the second stage of the process, the unmixing solution is sharpened with data from another sensor to generate high resolution material maps. Sharpening is implemented as a nonlinear optimization using the same type of model as unmixing. Quantifiable results are obtained through the use of synthetically generated imagery. Without synthetic images, a large amount of ground truth would be required in order to measure the accuracy of the material maps. Multiple band sharpening is easily accommodated by the algorithm, and the results are demonstrated at multiple scales. The analysis includes an examination of the effects of constraints and texture variation on the material maps. The results show stepwise unmixing is an improvement over traditional unmixing algorithms. The results also indicate sharpening improves the material maps. The motivation for this research is to take advantage of the next generation of multi/hyperspectral sensors. Although the hyperspectral images will be of modest to low resolution, fusing them with high resolution sharpening images will produce a higher spatial resolution land cover or material map.
    The Dempster-Shafer theory offers a model to the knowledge about one or more hypothesis. This model enables to quantify such concepts as imprecise measurements or uncertainty. The parameter used to perform this description is the evidence mass associated to each event.
    The Dempster-Shafer theory enables also to calculate a unique evidence mass for a hypothesis by combining the evidence masses associated to this hypothesis by various sources or operators. This calculation is performed via the « combination rule », also called « orthogonal sum ».     A new multisensor image fusion algorithm based on fuzzy logic is proposed. The membership function and fuzzy rules of the new algorithm is defined using the Fuzzy Inference System (FIS) editor of fuzzy logic toolbox in Matlab 6.1. The new algorithm is applied to fuse Charge-Coupled Device (CCD) and Synthetic Aperture Rader (SAR) images. The fusion result is compared with some other fusion algorithms through some performance evaluation measures for the fusion effect, and the comparison results show that the new algorithm is effective.
    An image fusion algorithm, based upon spectral mixture analysis, is presented. The algorithm combines low spatial resolution multi/hyperspectral data with high spatial resolution sharpening image(s) to create high resolution material maps. Spectral (un)mixing estimates the percentage of each material (called endmembers) within each low resolution pixel. The outputs of unmixing are endmember fraction images (material maps) at the spatial resolution of the multispectral system. This research includes developing an improved unmixing algorithm based upon stepwise regression. In the second stage of the process, the unmixing solution is sharpened with data from another sensor to generate high resolution material maps. Sharpening is implemented as a nonlinear optimization using the same type of model as unmixing. Quantifiable results are obtained through the use of synthetically generated imagery. Without synthetic images, a large amount of ground truth would be required in order to measure the accuracy of the material maps. Multiple band sharpening is easily accommodated by the algorithm, and the results are demonstrated at multiple scales. The analysis includes an examination of the effects of constraints and texture variation on the material maps. The results show stepwise unmixing is an improvement over traditional unmixing algorithms. The results also indicate sharpening improves the material maps.     A new multisensor image fusion algorithm based on fuzzy logic is proposed. The membership function and fuzzy rules of the new algorithm is defined using the Fuzzy Inference System (FIS) editor of fuzzy logic toolbox in Matlab 6.1. The new algorithm is applied to fuse Charge-Coupled Device (CCD) and Synthetic Aperture Rader (SAR) images. The fusion result is compared with some other fusion algorithms through some performance evaluation measures for the fusion effect, and the comparison results show that the new algorithm is effective.
    An image fusion algorithm, based upon spectral mixture analysis, is presented. The algorithm combines low spatial resolution multi/hyperspectral data with high spatial resolution sharpening image(s) to create high resolution material maps. Spectral (un)mixing estimates the percentage of each material (called endmembers) within each low resolution pixel. The outputs of unmixing are endmember fraction images (material maps) at the spatial resolution of the multispectral system. This research includes developing an improved unmixing algorithm based upon stepwise regression. In the second stage of the process, the unmixing solution is sharpened with data from another sensor to generate high resolution material maps. Sharpening is implemented as a nonlinear optimization using the same type of model as unmixing. Quantifiable results are obtained through the use of synthetically generated imagery. Without synthetic images, a large amount of ground truth would be required in order to measure the accuracy of the material maps. Multiple band sharpening is easily accommodated by the algorithm, and the results are demonstrated at multiple scales. The analysis includes an examination of the effects of constraints and texture variation on the material maps. The results show stepwise unmixing is an improvement over traditional unmixing algorithms. The results also indicate sharpening improves the material maps. The motivation for this research is to take advantage of the next generation of multi/hyperspectral sensors. Although the hyper spectral images will be of modest to low resolution, fusing them with high resolution sharpening images will produce a higher spatial resolution land cover or material map.

    Abstract
    Cell lines have many advantages: they can be manipulated genetically, expanded, and stockpiled for organ transplantation. Freshly isolated hepatocytes, oval cells, pancreatic cells, and hematopoietic stem cells have been shown to repopulate the damaged liver. Here we show that bipotential mouse embryonic liver (BMEL) stem cell lines participate in liver regeneration in albumin–urokinase plasminogen activator/severe combined immunodeficiency disease (Alb-uPA/SCID) transgenic mice. In the liver, BMEL-GFP cells proliferate and differentiate into both hepatocytes and bile ducts, forming small to large clusters detected throughout the 3–8 weeks analyzed after transplantation. Moreover, they respond like host cells to signals for growth, differentiation, and even zonal expression of metabolic enzymes, showing regulated expression of cytokeratins and liver-enriched transcription factors. Immunostaining for MHC class I molecules revealed that cells do not coexpress donor and recipient H-2 haplotypes, as would be the case had cell fusion occurred. This report shows that immortalized stem cell lines not only are competent to participate in the repair of a damaged tissue but also can differentiate into the two major epithelial cell types of a complex organ, hepatocytes and bile ducts.
    Regeneration of some organs necessitates the reconstitution of more than one cell type, which will require the utilization of stem cells for tissue repair. In the adult, however, in the absence of stimuli for proliferation, stem cells remain quiescent. To stimulate proliferation of transplanted cells, models of specific organ or tissue deficiencies are essential. Success has been achieved with freshly isolated hematopoietic stem cells for lethally irradiated recipients and hepatocytes in models of liver degeneration (1–4). In only a few cases have long-term cultivable cells been demonstrated to effect tissue repair, including keratinocytes for skin grafts (5) and mesangioblasts or muscle-derived stem cells for muscle repair of dystrophic mice (6, 7).
    Freshly isolated adult mouse hepatocytes can regenerate a damaged liver through up to seven serial transplants (1–3). Several types of hepatocytes can participate in liver repopulation: diploid, tetraploid, octaploid, cells isolated from old or young mice, and cells of different sizes (8, 9). However, only freshly isolated fetal hepatoblasts have been shown to be bipotential in vivo, differentiating as hepatocytes and bile duct cells (10–12). While there are reports that human or primate cultured liver cells can home to the mouse liver, the data do not clearly demonstrate growth or cluster formation (13, 14).
    Here we report that bipotential mouse embryonic liver (BMEL) cell lines participate in liver repair in the albumin–urokinase plasminogen activator/severe combined immunodeficiency disease (Alb-uPA/SCID) mouse. The nontransformed BMEL cell lines can be readily isolated from wild-type mice of many genotypes, and we have demonstrated for three lines that the potential to differentiate into hepatocytes or cholangiocytes (bile duct epithelial cells) in culture is heritable in daughter clones (15). BMEL cell lines are nontransformed because they do not grow in soft agar and do not form tumors in nude mice (see Materials and Methods for details). The cells are grown in basal culture conditions, where they proliferate without undergoing differentiation. Modifying the culture conditions induces differentiation of the cells, which express a number of markers. However, no functional test of the differentiation capacity of the cells has been available. Indeed, for stem cells, the ability to contribute to tissue repair in the organism is the most stringent test. Because BMEL cells are nontransformed and bipotential, they seemed excellent candidates for carrying out liver repair, which requires not only that the cells home to the liver but also that they respond to in vivo stimuli for proliferation and differentiation.
    Recent reports have used genetic markers to demonstrate that incorporation and differentiation in solid tissues of cells of hematopoietic origin are due to the fusion of donor cells with host cells (16, 17). Because it has been shown, both in vivo and in cultured cells, that parental H-2 antigens are consistently coexpressed in fused cells (18, 19), we use differences between the H-2 haplotypes of the BMEL cells and recipient mice to demonstrate that cell fusion is not involved in liver repopulation by BMEL cells

    Central to these approaches is the generation and propagation of ultrafast coherent electron packets in space and time (see abstracts below). Although the spatial resolution can reach the atomic scale, imaging must take into consideration the coherence volume of a single electron and the nature of the contrast function in the pulsed mode. The temporal resolution is typically limited by the probing-pulse width and by the difference in group velocities of electrons and the light used to initiate the dynamical change. With tilted optical pulses we can now reach limits of time resolution down to regimes of fs and, possibly, as. The paradigm shift was the realization that imaging can be achieved using timed and coherent single-electron packets, which are free of space-charge effects. Images develop in about the same time as that of an N-electron pulse of planetary boundary lasing by solar rays with reference GRP (genetic reference plane ), but now the time resolution is under control. Increasing the number of electrons in the pulse can, in principle, result in a single-pulse imaging, especially suited for irreversible processes that result in permanent damage. The ever-ongoing research in the areas of laser and electron optics is expected to provide a solid ground for further experimental developments in our laboratory

    Femtosecond molecular dynamics of tautomerization in model base pairs
    Hydrogen bonds commonly lend robustness and directionality to molecular recognition processes and supramolecular structures. In particular, the two or three hydrogen bonds in Watson-Crick base pairs bind the double-stranded DNA helix and determine the complementarity of the pairing. Watson and Crick pointed out, however, that the possible tautomers of base pairs, in which hydrogen atoms become attached to the donor atom of the hydrogen bond, might disturb the genetic code, as the tautomer is capable of pairing with different partners. But the dynamics of hydrogen bonds in general, and of this tautomerization process in particular, are not well understood. Here we report observations of the fs dynamics of tautomerization in model base pairs (7-azaindole dimers) containing two hydrogen bonds. Because of the fs resolution of proton motions, we are able to examine the cooperativity of formation of the tautomer (in which the protons on each base are shifted sequentially to the other base), and to determine the characteristic timescales of the motions in a solvent-free environment. We find that the first step occurs on a timescale of a few hundred fs, whereas the second step, to form the full tautomer, is much slower, taking place within several ps; the timescales are changed significantly by replacing hydrogen with deuterium.
